Additionally, he held multiple positions at Tufts University, including the director of graduate pharmacology from 1994 to 2000, associate professor of pharmacology, biochemistry and psychiatry from 1989 to 1994, director of medical pharmacology from 1985 to 1993, co-director of the medical pharmacology curriculum from 1983 to 1985, and assistant professor of biochemistry and pharmacology from 1983 to 1988. Previously, he served Yale University as a research associate with the faculty of clinical immunology from 1978 to 1983, and executive secretary for the university senate from 1976 to 1978. Dr. Theoharides began his career at Yale University as an assistant in research biology from 1968 to 1971, and an assistant in research in pharmacology from 1973 to 1978.

Before embarking on his professional path, Dr. Theoharides graduated from Anatolia College in Thessaloniki, Macedonia, Greece, earning a diploma, with honors, in 1968. He followed this accomplishment with a Bachelor of Arts cum laude and with Thesis Honors in biology and history of science and medicine in 1972, and a Master of Science in immunology in 1975, both at Yale University. He continued his academic efforts at Yale University, receiving a Master of Philosophy in endocrinology in 1975, as well as a Doctor of Philosophy in pharmacology in 1978, and a Doctor of Medicine in 1983. Dr. Theoharides concluded his studies with training in internal medicine at New England Medical Center from 1986 to 1993, as well as postgraduate coursework at Harvard University Kennedy School of Government, and at Tufts University Fletcher School of Law and Diplomacy.

Beyond his responsibilities within the field, Dr. Theoharides has contributed to numerous endeavors outside of his professional circles. He served as a special advisor for the Allergy Clinical Research Center at Attikon Hospital with the National and Kapodistrian University of Athens 2006-2008, and he has been the clinical pharmacologist with the Drug Formulary Commission for the Commonwealth of Massachusetts continuously since 1985. He has served as member of the Supreme Healthcare Board with the National Public Health Council for the Hellenic Republic from 2003 to 2008. As consultant to various companies, he participated in the development of a number of drugs (e.g. Ibuprofen, Cetirizine, Niaspan). Dr. Theoharides has had unique contributions in pharmacology and immunology, first demonstrating the ability of the tissue immune cells, the mast cells, to participate in inflammation by secreting their numerous mediators selectively, especially after stimulation by peptides secreted under stress, such as corticotropin-releasing hormone (CRH) and Substance P (SP). He then reported that mast cells are superstimulated by the combined action of SP and the cytokine interleukin IL-33 to secrete impressive amounts of the pro-inflammatory cytokines IL-1 and TNF He also showed that another peptide, neurotensin, is increased in the serum of children with autism spectrum disorder (ASD) and stimulates microglia to secrete IL-1 He also first showed that mast cell and microglia activation is potently inhibited by the novel natural flavonoid, methoxyluteolin, which offers promise for the treatment, in addition to allergies and asthma, of many difficult neuro-inflammatory conditions such as, ASD, myalgic encephalomyelitis/chronic fatigue syndrome, mastocytosis, multiple sclerosis and psoriasis. Dr. Theoharides takes pride in mentoring and has trained more than 200 medical, graduate and post-graduate students. He has been awarded numerous patents and trademarks.
